Microscopy and Measurements: Is it possible without a calibration slide?

I have an microscope camera, but no calibration slide. I also have access to the magnification values of the objective lenses. With a little math, would it be possible to calculate the length of objects under the microscope? Or should I just hold off until I get a calibration slide. (PS: I also have AmScope software that allows for measuring, but I have no clue how the calibration system works on that).

I measured all of my objectives with a calibration slide in case they were slightly different than labeled and remember one was a little different than I anticipated, so I’d say using a calibration slide is a good idea (especially if you ever end up in situations where a micron or two is significant). I have an extra if you’d like, you’re actually pretty close to me (I live in western Washington) so shipping or meeting up and dropping it off wouldn’t be an issue if that interests you!

I’m mostly interested in measuring the dimensions of fungal spores (very handy when identifying). I already ordered a calibration slide, for the sake of precision (the microns do matter!)

No worries! This has the same division size as mine, should be good for your purposes. I also use mine for fungal spores, though in my case it’s always ascospores.
