I’m unsure about my interpretation since I’m not much of a video game player, but I thought it might be a reference to the absorbing mushrooms in the Mario games. I may be wrong though, as I said not very familiar with the insider jokes of the gaming world.

The default of comedy is the subversion of expectation.

Aka you expect one thing, but its another…ha ha.

Pretty much the same joke, but a different direction of this.

The other classic ending to this cartoon is: (panel 3) the dog bites the standing man; (panel 4) the sitting man says “this isn’t my dog”.
